Ice Nucleation in Freeze - Tolerant Vertebrates

چکیده

The survival of freeze-tolerant vertebrates requires that ice nucleation in tissues be initiated at relatively high temperatures. Three current hypotheses for the initiation of freezing in these animals are: a) inoculation by contact with ice crystals in the external environment; b) nucleation by ice-nucleating bacteria associated with the animal; and c) nucleation by icenucleating proteins in the blood. The available evidence indicates that ice inoculation is the most important mechanism under natural conditions, although ice-nucleating bacteria harbored in the gut or skin during winter may function as an auxiliary system to promote crystallization, albeit at slightly lower temperatures. Despite the intriguing similarities between the blood icenucleating agents of some vertebrates and those of certain freeze-tolerant invertebrates, these agents seem unlikely to play an analogous, adaptive role in freeze tolerance.

Synchrotron X-Ray Visualisation of Ice Formation in Insects during Lethal and Non-Lethal Freezing

Although the biochemical correlates of freeze tolerance in insects are becoming well-known, the process of ice formation in vivo is subject to speculation. We used synchrotron x-rays to directly visualise real-time ice formation at 3.3 Hz in intact insects. We observed freezing in diapausing 3(rd) instar larvae of Chymomyza amoena (Diptera: Drosophilidae), which survive freezing if it occurs ab...

Production of Ice Nucleation Deficient (Ice-) Mutants of the Epiphytic Strains of Erwinia herbicola

To mutate the Ice Nucleation Active (INA) gene in Erwinia herbicola strains, Tn-5 transposon carried by Psup2021 plasmid was used. This plasmid was transferred to the bacterial cells by electroporation. Electrotransformation was carried out for 2.5 ms at 1800 v and 1 mm distance between the electrodes. Polymerase chain reaction was used for determination of presence or loss of INA gene, using a...

Animal ice-binding (antifreeze) proteins and glycolipids: an overview with emphasis on physiological function.

Ice-binding proteins (IBPs) assist in subzero tolerance of multiple cold-tolerant organisms: animals, plants, fungi, bacteria etc. IBPs include: (1) antifreeze proteins (AFPs) with high thermal hysteresis antifreeze activity; (2) low thermal hysteresis IBPs; and (3) ice-nucleating proteins (INPs). Several structurally different IBPs have evolved, even within related taxa. Proteins that produce ...

Supercooling and Freezing Tolerant Animals

Subzero temperatures may adversely affect animals by their direct lethal effects and by the damage caused by ice formation (Ramløv, 2000). Animals deal with the latter by three basic strategies. Freeze avoiding animals prevent ice formation in their bodies and supercool, keeping their body fluids liquid at temperatures below their melting point, but die if freezing occurs. In contrast, freezing...

Ice-Active Substances from the Infective Juveniles of the Freeze Tolerant Entomopathogenic Nematode, Steinernema feltiae

Steinernema feltiae is a moderately freezing tolerant nematode, that can withstand intracellular ice formation. We investigated recrystallization inhibition, thermal hysteresis and ice nucleation activities in the infective juveniles of S. feltiae. Both the splat cooling assay and optical recrystallometry indicate the presence of ice active substances that inhibit recrystallization in the nemat...
